Experiment Preparation of root tip squashes and study
5.2 of the stages of mitosis
Date ________________
Background The number of cells within an organism increases by mitosis and this is the
Information basis of growth in multicellular organisms. In plants, the roots continue to
grow. The root tips are good for studying the stages of mitosis.
Materials Microscope, slides, root tips provided by teacher, dissecting needle, oceto-orcin,
Required tissue paper
Procedure ΠPlace each root tip to a clean slide containing ! CAUTIONS
water drop.
Do not place too much
Dry it with a piece of soft tissue paper. It is material on a slide.
important to remove as much water as
possible.
Ž By using a dissecting needle, chop the root tip thoroughly and spread it
on the slide.
Add 1 drop of oceto-orcin stain on the root tips and stay for 3 minutes.
Remove the extra stain by filter paper.
‘ Place a cover slip gently by avoiding air bubbles.
’ Observe the slide under the microscope at low and high powers.
