7.2 (CS) of a leaf through observations under
the microscope

Background In cross section of a leaf first layer is upper epidermis, covered with cuticle.
Information Below the upper epidermis is mesophyll tissue, differentiated into palisade and
spongy tissues. Vascular bundles are present in spongy tissue. The bottom layer
is the lower epidermis which contains most of the stomata.

Materials Prepared slide of the CS of leaves, microscope

Procedure Œ Examine the prepared slide of CS leaf ! CAUTIONS
Observations under low and high powers of microscope.
• Handle the slides
Draw a diagram of the observed slide. carefully.

• Clean the slide,
ocular lens, and
objective of
microscope before
observing.
